- Contamination / Negative Markers
Canonical Markers
Validated antibodies and ELISA kits against the tetraspanins CD9, CD63, CD81, CD82, and CD151, ESCRT/biogenesis proteins TSG101, ALIX (PDCD6IP), and Syntenin (SDCBP), the lipid raft protein Flotillin-1 (FLOT1), and the cargo chaperones HSP70 (HSPA8) and HSP90 — for Western blot, ELISA, flow cytometry, and mass spectrometry.
Biogenesis & Trafficking Markers
Antibodies against the regulators that drive EV cargo sorting and release: small GTPases RhoA and RAB27A/B, RAB11, and RAB35, ESCRT machinery components VPS4A/B, CHMP4B, VPS37B, and MVB12A/B, and the lipid-mediated biogenesis enzyme SMPD2 — essential for dissecting exosome vs. microvesicle biogenesis pathways.
Cell-of-Origin / Lineage Markers
Trace disease-relevant EV populations back to their parent cell: CD14, CD11b, and ICAM-1/CD54 (monocyte/immune), S100A9, TMEM119, and P2RY12 (microglia), GAD1, GAD2, CHAT, GPM6A, and L1CAM (neuron), GFAP (astrocyte), SLC12A1 (kidney/epithelial), and EpCAM and CD90/Thy1 (epithelial, tumor, and stem/mesenchymal EVs) — key for liquid biopsy and biomarker discovery.
Functional, Stress & Tumor-Associated Cargo Markers
Antibodies and ELISA kits for stress-responsive and tumor-associated cargo Annexin A2 (ANXA2), Galectin-3 (LGALS3), AHNAK, AHNAK2, and MFG-E8, immunomodulatory molecules PD-L1 and TGF-β, and metabolic enzymes FASN, ITIH4, IDH1/2, UQCRC1/2, and LRRC15 that shape recipient-cell behavior.
Contamination / Negative Markers
Confirm sample purity per MISEV2018 by demonstrating the absence of organelle-derived contaminants: Calnexin (CANX) and HSP90B1/GRP94 (endoplasmic reticulum), GM130/GOLGA2 (Golgi), Cytochrome c (CYCS) and ATP5A1 (mitochondria), Lamin B1 (nucleus), and Actin (ACTB) and Vimentin (VIM) (cytoskeleton) — via Western blot or proteomics.

Published Evaluation
Rapid EV Detection, Verified in an Ovarian Cancer Model
Researchers at Nagoya University evaluated the Exorapid-qIC™ Immunochromatographic Kit using small EVs (sEVs) secreted by the human ovarian adenocarcinoma cell line CAOV3.
CD9 and CD63 test lines were both detected in under one hour, with visible signal once particle counts exceeded 1×10⁸ particles/test by NTA.
CD9 and CD63 detection trends correlated with previously published CAOV3 data.
Comparing four independently prepared sEV lots revealed batch-to-batch variability that NTA analysis alone did not capture — underscoring the value of multi-method EV evaluation.
"Using this kit, sEVs secreted by the human ovarian cancer cell line CAOV3 could be easily detected in a short time — though variability between preparations highlights the importance of evaluating sEVs using multiple methods."
