Osteoclast cells (human) are bone marrow mononuclear cell-derived osteoclast precursor cells (1.5x106cells/vial). For guaranteed optimal recovery and differentiation we recommend Osteoclast Wash Medium (PMC-OSCMW) and CSF/RANKL-containing Osteoclast Culture Medium (for human) (PMC-OSCMHB). Together, these products are useful for studies of osteoclast formation, bone resorption, osteoporosis, rheumatoid arthritis, Paget's disease and others.
Bone mass is controlled by the dynamic interplay of bone-forming osteoblasts and bone-degrading osteoclasts. In recent years, a system has been established that induces bone marrow cells to differentiate into osteoclasts using M-CSF (Macrophage Colony Stimulating Factor) and RANKL (Receptor Activator of NF kappa B Ligand).

Upon receipt of dry ice-packaged shipment directly from Japan, either immediately store in liquid nitrogen or introduce cells directly into culture. Biosafety level 2 containment precautions should be used when handling this product. For guaranteed optimal recovery and differentiation we recommend Osteoclast Wash Medium (PMC-OSCMW) and CSF/RANKL-containing Osteoclast Culture Medium (for human) (PMC-OSCMHB). Together, these products are useful for studies of osteoclast formation, bone resorption, osteoporosis, rheumatoid arthritis, Paget's disease and others.
Product Warranty
When cells have been properly stored in liquid nitrogen and cultured according to instructions using recommended media and reagents, we guarantee against poor growth at the initiation of culture. The warranty period is [within 6 months after receiving the product]. Warranty is not applicable if changes are made to the media or its usage, or if refrozen cells are utilized.

What is the composition of Rat Osteoclast Culture Medium (Cat. No. PMC-OSCMR), Mouse Osteoclast Culture Medium (Cat. No. PMC-OSCMM) and Human Osteoclast Culture Medium (Cat. No. PMC-OSCMHB)? These culture media consist of α-MEM, 10% FBS, M-CSF, RANKL and Penicillin-Streptomycin.
How can I observe pits on dentine slices when using Rat Osteoclast Cells (Cat. No. PMC-OSC12C), Mouse Osteoclast Cells (Cat. No. PMC-OSC14C) or Human Osteoclast Cells (Cat. No. PMC-OSC15C) in a pit formation assay? After staining the samples with hematoxylin, you can observe the pits on dentine slices under a stereomicroscope.
How long was Human Osteoclast Cells (Cat. No. PMC-OSC15C) cultured to generate the pits shown in Figure 4 of the datasheet? The pits on the ivory slice were formed after 2 weeks of culture.
Can I passage Rat Osteoclast Cells (Cat. No. PMC-OSC12C), Mouse Osteoclast Cells (Cat. No. PMC-OSC14C) or Human Osteoclast Cells (Cat. No. PMC-OSC15C)? Generally, it is difficult to detach osteoclasts from culture dishes using trypsin treatment. Known methods for passaging mature osteoclasts include physically detaching them with a cell scraper or culturing them on collagen gel plates beforehand. However, please note that both methods generally result in low recovery rates for viable cells.
Can I use Rat Osteoclast Culture Medium (Cat. No. PMC-OSCMR), Mouse Osteoclast Culture Medium (Cat. No. PMC-OSCMM) and Human Osteoclast Culture Medium (Cat. No. PMC-OSCMHB) after refreezing? If you plan to use these culture media in multiple experiments, aliquot them into 10 mL tubes after thawing and store the aliquots at -20°C or below. Repeated freeze-thaw cycles should be avoided because they may reduce the activity of M-CSF and RANKL. Once thawed, each aliquot should be used immediately.
How should I culture Rat Osteoclast Cells (Cat. No. PMC-OSC12C), Mouse Osteoclast Cells (Cat. No. PMC-OSC14C) or Human Osteoclast Cells (Cat. No. PMC-OSC15C) to prevent them from differentiating into osteoclasts? We recommend using these culture media without RANKL. To avoid culture variations caused by different serum lots, please use Osteoclast Wash Medium (Cat. No. PMC-OSCMW). Also, you should add M-CSF to the washing medium.
