Background
Arraying cells is an important step for single cell analysis. For example, cell overlap is unfavourable for single cell imaging and isolation. Microcavity arrays are used to capture single cells. In general, the distribution of cells in a cavity is by sedimentation and the single cell rate follows a Poisson distribution. Its single cell capture rate is relatively low, especially when loading cells in higher quantities.

What nanowell sizes are available for SIEVEWELL™ Slide (Cat. No. TOK-SWS-2001-5 / TOK-SWS-5001-5)? We offer two nanowell sizes: 20 µm (Cat. No. TOK-SWS-2001-5) and 50 µm (Cat. No. TOK-SWS-5001-5).
Is SIEVEWELL™ Slide (Cat. No. TOK-SWS-2001-5 / TOK-SWS-5001-5) compatible with most inverted microscopes? SIEVEWELL™ Slide is compatible with most inverted microscopes that have a standard stage or frame.
Is SIEVEWELL™ Slide (Cat. No. TOK-SWS-2001-5 / TOK-SWS-5001-5) reusable? No. SIEVEWELL™ Slide is intended for single use only and is not reusable.
Can I load cells onto SIEVEWELL™ Slide (Cat. No. TOK-SWS-2001-5 / TOK-SWS-5001-5) and culture them directly? Yes, you can culture cells directly on this SIEVEWELL™ Slide. The slide includes a protective lid for cell culture.
Do cells grow directly in the nanowells of SIEVEWELL™ Slide (Cat. No. TOK-SWS-2001-5 / TOK-SWS-5001-5)?
Yes. Depending on the cell type, some cells attach within or around the nanowells, while others remain in suspension without adhering.

Can I perform live-cell imaging with SIEVEWELL™ Slide (Cat. No. TOK-SWS-2001-5 / TOK-SWS-5001-5)?
Yes. Live-cell imaging can be performed with SIEVEWELL™ Slide.
You can monitor live cells over time and subsequently recover viable single cells or spheroids.

Can I retrieve cells from SIEVEWELL™ Slide (Cat. No. TOK-SWS-2001-5 / TOK-SWS-5001-5)? Yes. Cells can be retrieved by gently flushing medium or DPBS through the side port several times.
Which solvents and chemicals are compatible with SIEVEWELL™ Slide (Cat. No. TOK-SWS-2001-5 / TOK-SWS-5001-5)?
Acetone is not compatible and will damage SIEVEWELL™ Slide.
Please note that chemical compatibility with other solvents may vary depending on physical conditions such as temperature, strain, or when used for extended periods of time. We recommend testing the chemical compatibility with your specific solvent using a trial sample prior to your experiments. Please contact us to request a trial sample.
Is high-resolution microscopy (40×, 60×, 100×) possible with SIEVEWELL™ Slide (Cat. No. TOK-SWS-2001-5 / TOK-SWS-5001-5)?
High-resolution microscopy (using 40×, 60×, or 100× objectives) is not suitable for live-cell imaging because the thickness of the device bottom plate prevents proper focusing.
For fixed cells, confocal microscopy can be achieved by disassembling the device and mounting the sample between coverslips.
