Product Description

Pink Flamindo is a single-color, intensity-change-type red fluorescent protein sensor that visualizes the dynamics of cyclic adenosine monophosphate (cAMP) inside cells. The cAMP-binding domain of Epac1, a protein with affinity for cAMP, is genetically fused near the chromophore of the red fluorescent protein mApple. Furthermore, the amino acid sequence of the linker region connecting the two proteins has been optimized. The dissociation constant (Kd) for cAMP is 7.2 µM. Real-time observation of cAMP production induced by photoactivated adenylyl cyclase (PAC) can be achieved under blue light illumination.

Red Glifon 300 and Green Glifon 4000 are fluorescent protein-based molecular sensors whose fluorescence intensity changes in response to glucose concentration.
Red Glifon 300 consists of a split red fluorescent protein (mApple) fused with the glucose-binding domain of MglB, and its EC₅₀ value for glucose is 320 µM.
Green Glifon 4000 consists of a split green fluorescent protein (Citrine) fused with the glucose-binding domain of MglB, and its EC₅₀ value for glucose is 4000 µM.

Changes in fluorescence intensity enable visualization of intracellular glucose dynamics, allowing spatial and hierarchical observation of cellular glucose metabolism. By combining these sensors with optogenetic or in vivo imaging techniques, and using systems such as image-splitting optics, simultaneous visualization with other sensors is possible.
