What are glycan-immobilized gold nanoparticles (SGNPs)?
SGNPs (Sugar chain-immobilized Gold Nano-Particles) are gold particles modified with sugar chains, approximately 10 nm (10 x 10⁻⁹ m) in size.
They selectively bind to sugar-binding proteins such as lectins, and if the protein is polyvalent, they exhibit aggregation and precipitation in solution, allowing for visual observation of their binding behavior. Furthermore, SGNP solutions have absorption around 530 nm (resulting in a red wine-like color), enabling quantification of aggregation levels and confirmation of binding by dot blotting.
Examples of experiments using SGNPs
1. SGNP-protein aggregation experiment
SGNPs can be used to investigate their binding activity to proteins.
2. SGNP-protein aggregation inhibition experiment
An aggregation inhibition experiment can be conducted by adding an inhibitor.
3. Dot blotting
A small amount of protein is spotted onto a membrane and detected using SGNPs.
4. Isolation and identification of proteins from crude extracts
By utilizing the properties of aggregation and dissociation, the target protein can be isolated from a mixed solution.
