Antibodies from Cosmo Bio to Humanin (HN) and potent analog HNG (S14G) accelerate research in cognitive aging and Alzheimer’s disease

Alzheimer’s disease (AD) is a neurodegenerative disease characterized by impairment of cognitive function, death of neuronal cells, the presence of senile plaques and neurofilament tangles. Humanin was originally identified as a mitochondrial genome-encoded 24-residue, secreted peptide possessing potent neuroprotective activity against neuronal cell death from mutant forms of the three known familial AD genes APP, PS1, and PS2, as well as from treatment with Amyloid-beta peptide 1-43 (1). In animal models Humanin has been shown to powerfully protect from cognitive declines associated with both AD and normal aging (2). Neuroprotective effects of Humanin are thought to be exerted extracellularly through interaction with cell surface receptors, including the Formyl Peptide Receptor 2 and tripartite IL6-receptor-related complex comprising CNTF Receptor-Alpha/WSX-1/gp130 (3, 4). Humanin has also been shown to act intracellularly to suppress apoptosis by binding the pro-apoptotic protein Bax (5). Humanin neuroprotective activity is potentiated by binding Insulin-like growth factor-binding protein-3 (6). Intracellular Humanin levels may be regulated by binding the E3 ubiquitin-protein ligase Trim11 (7).

To our knowledge, there is only one source of an antibody to the Humanin analog S14G (also known as HNG) which in multiple assays of neurodegneration is 1000-fold more biologically potent than Humanin itself. Our collection of anti-Humanin antibodies (including anti-S14G) and anti-Humanin ELISA kit are useful in studies of AD as well as of other diseases (such as Type 2 diabetes) where amyloid and amyloid-like deposits cause pathology. In addition, we carry a broad range of useful downstream assays and reagents for study of neurodegenerative diseases such as AD.
