 Brown adipocyte culture kit (rat)

The brown adipocyte culture kit is a kit for culturing brown adipocytes related to obesity and diabetes. Using this culture system, lipid metabolism experiments of brown adipocytes, thermal energy release experiments, functional elucidation of brown adipocytes, screening of new β 3 agonists, etc. are possible.

Background

Brown adipose tissue has the special function of storing excess energy as fat, and at the same time releasing the fat energy directly to the outside of the body as heat. In addition, the β action of noradrenaline secreted from the sympathetic nerves contributes to the automatic regulation of energy consumption.
This culture kit (Part No.: BAT01, BAT02) is shipped after primary culture of brown adipose precursor cells from neonatal rat brown adipose tissue and proliferation to semi-confluence. If the primary cultured cells at the time of arrival, or the cells subcultured in an appropriate culture dish or multiplate are cultured according to the culture method, they will gradually differentiate into brown adipocytes.
Product number: BAT10C is a frozen primary brown adipose precursor cell collected from mature rat brown adipose tissue. It is possible to culture in either a medium that does not contain a forced differentiation agent (product number: BATFM) or a medium set that contains a forced differentiation agent (product numbers: BATGM, BATDM, BATMM).

Cultured Cells : These cells are delivered in culture vessels (adherent cells or suspension cells). Since it cannot be stored in that state, please take it out immediately after delivery and immediately start culturing in a CO2 incubator.
Frozen Cells : The cells are shipped frozen in dry ice packaging. You can take it out immediately after delivery and start culturing immediately, or you can freeze it at the temperature specified in the instruction manual.

We investigated the activation of UCP-1 in brown adipocytes (BAT10C, BATFM) and 3T3-L1 cells.
Cells with sufficient accumulation of lipid droplets were stimulated with noradrenaline (exposure for 6 hours), and the expression level of UCP-1 was analyzed by RT-PCR.
As a result, the UCP-1 gene was elevated in noradrenaline-stimulated brown adipocytes,
but he could not detect the UCP-1 gene in 3T3-L1 cells even with noradrenaline stimulation.
